The 307.9D1 monoclonal antibody reacts with mouse Repulsive Guidance Molecule b (RGMb), a member of the RGM family which consists of RGMa, RGMb, and RGMc/hemojuvelin. RGMs are GPI-anchored membrane proteins that bind bone morphogenetic proteins (BMPs) and neogenin. BMPs have diverse roles in many processes, including cell proliferation, differentiation, and apoptosis. RGMb has a well-elucidated role in neural development, with RGMb knock-out mice dying 2-3 weeks after birth. RGMb is also expressed by macrophages and can regulates IL-6 expression. Recently, it has been shown that PD-L2 also binds to RGMb and that blocking the RGMbāPD-L2 interaction impairs the development of respiratory tolerance but blocking the PD-1āPD-L2 interaction does not. The 307.9D1 antibody has been shown to block the binding of RGMb to PD-L2 and BMP-2/4.Specifications
